However,theanalysisofN-glycanstructuresisstill challengingduetothestructuraldiversityandheterogeneityof glycoproteinsandtheirN-glycans.Typically,theN-glycansare releasedfromtheproteinbackbonebyenzymaticcleavage, followedbyderivatizationandchromatographicseparationbefore massspectrometryanalysis.However,thisapproachusuallyresults inthelossofinformationabouttheintactN-glycanstructures, especiallyforlargeglycoproteinswithcomplexN-glycanpatterns. Therefore,alternativemethodsforthecomprehensiveanalysisof N-glycanstructuresofglycoproteinsareneeded. Ultrafiltrationmembrane-basedmethodshaveemergedas promisingtoolsforglycananalysisinrecentyears.The ultrafiltrationmembraneshavedefinedporesizesthatcan separatemacromoleculesbasedontheirmolecularweight, allowingfortheselectiveenrichmentofglycanchainsfromthe proteinmixture.Inthisstudy,wedevelopedanultrafiltration membrane-basedmethodfortheenrichmentandmass spectrometryanalysisoftheentireN-linkedglycanchainsof glycoproteins.
