It is rich in avariety of amino acids, which contribute to its therapeutic properties. This study aimed to investigate the optimal extraction process for complex amino acids from donkey-hide gelatin. Different extraction techniques were compared, including acid hydrolysis, enzymatic hydrolysis, and microwave-assisted extraction. The effects of various factors, such as temperature, extraction time, and enzyme concentration, were evaluated. The results demonstrate that enzymatic hydrolysis at a moderate temperature and enzyme concentration is the most effective method for extracting complex amino acids from donkey-hide gelatin. This research provides valuable insights into the production of high-quality Ejiao products. 1. Introduction Donkey-hide gelatin, atraditional Chinese medicine, has been used for centuries due to its various health benefits. It is primarily composed of collagen, aprotein that can be hydrolyzed into various amino acids during the extraction process. These amino acids play acrucial role in the therapeutic effects of donkey-hide gelatin. However, the extraction of complex amino acids from donkey-hide gelatin remains achallenge. Therefore, it is important to explore the optimal extraction process to maximize the yield and quality of amino acids. 2. Methods 2.1 Sample Preparation Donkey-hide gelatin was collected and dried. The dried gelatin was ground into afine powder for further analysis. 2.2 Extraction Techniques Three extraction techniques were compared: acid hydrolysis, enzymatic hydrolysis, and microwave-assisted extraction. In acid hydrolysis, the gelatin powder was mixed with hydrochloric acid (HCl) at various concentrations and subjected to hydrolysis under different temperatures and times.
