养猪发酵床中净水芽孢杆菌的分离及其固体发酵研究 摘要： 本研究旨在从养猪发酵床中分离并鉴定净水芽孢杆菌，并对其进行 固体发酵的研究。通过在发酵床样品中筛选，得到一株菌株，经过16S rRNA基因序列分析和形态学检测并鉴定为净水芽孢杆菌。在固体发酵的 实验中，探究了pH、温度、初始菌液浓度和底物比例等对净水芽孢杆菌 生长和产生发酵产物的影响。结果表明，在初始pH为7.0、温度为 30℃、初始菌液浓度为10%、底物比例为1:1的条件下，净水芽孢杆菌 可以发出最大的酶活力和生长率，生产的酶活力分别为5.3U/mL和3.2 U/mL。本研究结果为利用净水芽孢杆菌在固体发酵中产生高效复合酶提 供了理论依据。 关键词：养猪发酵床；净水芽孢杆菌；16SrRNA；固体发酵；复合 酶 Abstract: TheaimofthisstudywastoisolateandidentifyBacillus licheniformisfrompig-breedingfermentationbed.Furthermore, thesolid-statefermentationofBacilluslicheniformiswas investigated.Throughscreeningsamplesfromafermentationbed, astrainwasisolatedandidentifiedasBacilluslicheniformis through16SrRNAgenesequenceanalysisandmorphological detection.Intheexperimentsofsolid-statefermentation,the effectsofpH,temperature,initialbacterialliquidconcentration andsubstrateratioonthegrowthandproductionoffermented productswerestudied.TheresultsshowedthatBacillus licheniformiscanproducethelargestenzymeactivityandgrowth rateundertheconditionsofinitialpH7.0,temperature30℃,initial bacterialliquidconcentration10%,andsubstrateratio1:1.The enzymeactivitiesproducedwere5.3U/mLand3.2U/mL, respectively.Ourstudyprovidesatheoreticalbasisfortheuseof Bacilluslicheniformistoproduceefficientcomplexenzymes throughsolid-statefermentation.
