人突变型p53和EB病毒LMP1转基因小 鼠的建立 作者：何迎春，田道法，卢芳国，毛积芳【关键词】鼻咽癌 前病变 Constructionoftransgenicmicecontaininghumanmutant p53andEBviruslatentmembraneprotein1 【Abstract】AIM:Toestablishbitransgenicmicemodels containingmutationp53geneandEpsteinBarr(EB)virus latentmembraneprotein1gene（LMP1）andtoprovide reliabletoolsforstudyingthereciprocityofmutantp53 withLMP1inthetumorogenesisprocessofnasopharyngeal precancerosis.METHODS:Molecularcloningtechniqueswere usedtoconstructtherecombinantplasmidpLMP1p53mt containingmutantp53geneandEBvirusLMP1gene.Linear recombinantplasmidpLMP1p53mtwastransfectedintomouse androgenesisofgermcellsbymicroinjectionandthen survivalgermcellstreatedwereplantedintofallopian tubesofartificialpregnantfemalemice.Threeweekfilial generationmicewerescreenedbyPCRandfurtheridentified bysouthernblot,andthehistopathologicalcharacteristics
