丙戊酸钠抑制HL-60细胞增殖并诱导其凋亡与分化_医学论文作者：韩冬 梅，陈协群，梁蓉，董宝侠，尹文 【摘要】为进一步研究丙戊酸钠（sodiumvalproate；VPA）对白血病细胞 HL-60的增殖、凋亡和分化的影响，采用MTT法检测不同浓度的VPA对HL-60 细胞增殖的作用；细胞化学染色法观察药物作用后细胞形态的变化；NBT还原 实验结合形态学作为观测细胞分化指标；应用流式细胞术检测不同浓度药物作 用后细胞周期的变化及凋亡细胞的比例。结果显示：VPA能明显抑制HL-60的 增殖；经2mmol/LVPA处理24-48小时后，HL-60出现胞浆空泡、核固缩、核 碎裂及凋亡小体；AnnexinV阳性的早期凋亡细胞比例由2.9%升高到17.1%； 流式细胞术检测出明显的亚二倍体峰；G1期细胞由51.1%增加至84.6%，S期细 胞由37.9%减低至14.4%。0.25mmol/LVPA作用1周后，促进HL-60细胞向中 幼粒、晚幼粒阶段分化，NBT阳性细胞百分率为（47±2）%。结论：VPA能明显 抑制白血病细胞HL-60的增殖，并诱导其分化及凋亡。 【关键词】丙戊酸钠 InhibitionofProliferationandInductionofApoptosisand DifferentiationofLeukemicCellLineHL-60bySodiumValproate AbstractToinvestigatetheinfluenceofsodiumvalproate(VPA) onproliferation，apoptosisanddifferentiationofHL-60cellline， effectofVPAinvariousconcentrationsonproliferationofHL-60 cellswasdetectedbyMMT；Wright-Giemsastainingwasperformedto observethemorphologicchangesofHL-60cells；NBTexperimentwas usedtotestthedifferentiationofHL-60cells；flowcytometrywas usedtoobservecellcyclesandanalyzetheapoptosis.Theresults indicatedthatthechangesofthegrowthcurveshowedinhibitionof proliferationofHL-60cells.Aftera24-48hoursculturewith2 mmol/LVPA，thecellsexhibitednuclearshrinkage，pyknosis fragmentationandappearenceofapoptosisbodies.Thepercentageof theannexinV+/PI-cellswhichwereapoptoticincreasedfrom2.9%to 17.1%；hypodiploidpeakwasobserved；thepercentageofHL-60cells inG1phaseincreasedfrom51.1%to84.6%andthecellsinSphase decreasedfrom37.9%to14.4%.Afteraweekculturewith0.25mmol/L VPA，thecellsexhibitedcharacteristicsofdifferentiation.The percentageofNBTpositivecellswas(47±2)%.Itisconcludedthat VPAcaninhibittheproliferationofHL-60cellswhileinducing differentiationandapoptosisofthesecells.Themechanismneedsto befurtherstudied.
