低氧对小鼠卵母细胞体外成熟及卵丘颗粒细胞转录组 miRNA 和表达谱变化的影响 Title: Effects of Hypoxia on In Vitro Maturation of Mouse Oocytes and the Transcriptional and miRNA Expression Profiles of Cumulus Granulosa Cells Abstract: Hypoxia, acondition characterized by low oxygen levels, has been shown to have various effects on cellular function and development. This study aimed to investigate the impact of hypoxia on the in vitro maturation (IVM) of mouse oocytes and the transcriptional and miRNA expression profiles of cumulus granulosa cells (CGCs). Our findings suggest significant changes in oocyte maturation and gene expression patterns in CGCs under hypoxic conditions. These results provide valuable insights into the potential role of hypoxia in the regulation of oocyte maturation and cumulus cell function. Introduction: Oocyte maturation is acritical step in mammalian reproductive physiology and is tightly regulated by complex interactions between the oocyte and its supporting cumulus granulosa cells (CGCs). CGCs are responsible for the synthesis and secretion of various molecules and provide essential support for oocyte maturation. Recent studies have shown that environmental factors, such as hypoxia, can significantly influence oocyte maturation and the functions of CGCs. However, the precise molecular mechanisms underlying these effects are not well understood. Methods: 1. Experimental design: -Control group: Oocytes and CGCs cultured under normal oxygen conditions (21% O2). -Hypoxia group: Oocytes and CGCs cultured under reduced oxygen conditions (5% O2). 2. Oocyte collection and culture: -Oocytes were collected from superovulated female mice and
