探究参与抗病毒天然免疫信号通路的新分子的开题报 告 Title:Investigationofnovelmoleculesinvolvedinanti-viral innateimmunesignalingpathways Introduction: Theinnateimmunesystemplaysacrucialroleinprotecting organismsfrompathogenicinfections,andactivationoftheinnate immunesystemisinitiatedbypathogen-associatedmolecular patterns(PAMPs)thataredetectedbypatternrecognition receptors(PRRs).UpondetectionofPAMPsbyPRRs,an intracellularsignalingcascadeisactivatedwhichultimatelyleads totheinductionoftypeIinterferon(IFN)andproinflammatory cytokines,andtheinductionofapoptosis.Amongthesepathways, theRIG-I-likereceptors(RLRs)andtoll-likereceptors(TLRs)are importanttypesofPRRsthatareinvolvedinanti-viralinnate immunesignaling. Recentworkhasrevealedthatanumberofmoleculesare involvedintheRLRandTLRpathways,butmanyofthese moleculesarenotyetfullyunderstood.Assuch,thisprojectseeks toinvestigatenovelmoleculesinvolvedinanti-viralinnate immunesignalingpathways,withafocusontheRLRandTLR pathways. Objectives: 1.Toscreenalibraryofgeneknockdowncellsforthosethat aredefectiveinIFNandcytokineinductionafterchallengewith viralPAMPs. 2.Toidentifythemoleculesalteredintheabovescreening, usingRNAseqandproteomicapproaches,andinvestigatetheir roleinanti-viralinnateimmunesignalingpathways. 3.Toinvestigatethesignalingpathwaysthroughwhichthese moleculesfunctionusingbiochemical,genetic,andcellbiological approaches.
